Molina's side are currently atop the Group F standings with seven points, with Johor one point adrift.
However, Kitchee will not be guaranteed a spot in the knockout stage of the AFC Cup even if they beat Johor.
"It's going to be tough, as they will have the support of thousands of fans," Molina told the South China Morning Post.
But, Molina conceded that his side's chances of defeating Johor have been boosted by the fact that they have a full-strength squad available.
"We are happy to have our best players here, including captain Recio Fernando," he said.
Meanwhile, Johor coach Bojan Hodak believes that his side can overtake Kitchee atop the points table, with their home ground advantage key.
However, Hodak confirmed that goalkeeper Farizal Marlias will miss the match as he suffered a concussion during Johor's Malaysia Super League game against Singapore Lions XII on Saturday.
"We are playing in front of our fans in our full stadium where we have a perfect record in AFC competitions this year," Hodak told the official AFC website. "This is an important game because the team that wins will go top of the group, so I hope we can deliver at home.
"Lucho (Figueroa) will play because he has recovered from injury, but we will have to see how long he can last.
"We don't have any other injury, except Farizal Marlias because he was concussed after the last league game against Lions XII and will be out for a few days."
